Primary: nontreponemal serologic test (RPR or VDRL), Darkfield microspcopy of the lesion to see the spirochete. Direct antigen test ( MHA-TP or F TA-ABS) to confirm.

Secondary:Clinical symptoms plus positive serologic tests

Tertiary: I am not too sure
