High TSH level is one of very common and hidden cause of Hyperprolactinemia so have to be ruled out.

Progestine challege test will let u know that there is no ovulation, but u cant know the reason behind it.

Sachin, don't be confused.

Let me tell you, the basic physiology of menstruation is soly bec of oestrogen in proliferative and progesterone in secretary phase , right?

All other hypothalamic, pituitary, hormonal axis affect menstruation via low estrogen, an ovulation with low progesterone,

So, whatever the cause, the basic main cause is low estrogen and low progesterone causing amenorrhoea. Except out flow obstruction.

So , the best test should be...for these two. Ofcourse we check TSh, prolactin. But not the best next step for amoenorrhoea.
