From a collection of mutated consensus sequences and a preparation containing transcription factor from a cancer cell line, you propose to find the better binding sequences by passing a mixture of the transcription factor and the collection of sequences through a gel-filtration chromatography column of a type that excludes the transcription factor in question from retention. Each of the pieces of consensus sequence has a radioactive tag at one of the ends. The transcription factor is going to be washed out of the gel no matter what. However, if it binds onto one of sequences, the attached radioactive tag goes out with it in the fraction.
