With Explaination pease........
which of the following is the major disadvantage of naloxone as a treatment for opioid overdose??

a) as a partial agonist,it may produce depression of the central nervous system
b) It does not antagonize the effect of heroin
c) It has a low theraputic index
d) It has a shorter duration of action than most opioids
e) Tolerance develops to its antagonistic effect

--Naloxone is a μ-opioid receptor competitive antagonis...hence choice A is out
--Naloxone is antagonist to heroin---hence B is out
--Naloxone possesses a high therapeutic index, but it may provoke withdrawal signs and symptoms(seizures....)

--Naloxone IV duration of action is 40 -75 minutes...whereas many opiods produce clinical effects that last 3 to 6 hours...hence naloxone is used only for acute opioid toxicity...whereas naltrexone has longer duration of action and is used for LONG TERM DETOXIFICATION THERAPY...

--I never came across with tolerance to naloxone...

hence my answer is DDD

Naloxone has a shorter duration of action...and is combined with buprinorphine subligually then wuld have a 3day half life.
Gnerally nalotrexone is guiven bcos of its longer half life
